Anomalous Angiogenesis in Retina

چکیده
Age-related macular degeneration (AMD) may cause severe loss of vision or blindness, particularly in elderly people. Exudative AMD is characterized by the angiogenesis blood vessels growing from underneath macula, crossing blood–retina barrier (which comprises Bruch’s membrane (BM) and retinal pigmentation epithelium (RPE)), leaking fluid into retina knocking off photoreceptors. Here, we simulate a computational model choroid via cellular Potts model, as well BM, RPE cells, drusen deposits Our results indicate that improving require fixing impaired lateral adhesion between cells with diminishing Vessel Endothelial Growth Factor (VEGF) Jagged proteins affect Notch signaling pathway. numerical simulations suggest anti-VEGF anti-Jagged therapies could temporarily halt exudative while addressing adhesion, which be more effective over longer time-span.

Functional analyses of TNFR2 in physiological and pathological retina angiogenesis.
PURPOSE To determine the function of tumor necrosis factor receptor-2 (TNFR2) in retinal development and ischemia-induced revascularization in an oxygen-induced retinopathy (OIR) model. METHODS Mice with a global deletion of TNFR2 (TNFR2-KO) or with a vascular endothelial cell (EC)-specific TNFR2 transgene (TNFR2-TG) were compared to wild-type C57BL/6 mice (WT). Retinal vasculature developmen...
متن کاملProlactins are natural inhibitors of angiogenesis in the retina.
PURPOSE Disruption of the normally antiangiogenic environment of the retina leads to aberrant angiogenesis, the major cause of vision loss throughout the world. Prolactin (PRL), the hormone originally associated with milk production, can be proteolytically processed to 16K-PRL, a 16 kDa N-terminal PRL fragment with potent antiangiogenic and vasoconstrictive actions. This study was conducted to ...
متن کاملNeurons Limit Angiogenesis by Titrating VEGF in Retina
Vascular and nervous systems, two major networks in mammalian bodies, show a high degree of anatomical parallelism and functional crosstalk. During development, neurons guide and attract blood vessels, and consequently this parallelism is established. Here, we identified a noncanonical neurovascular interaction in eye development and disease. VEGFR2, a critical endothelial receptor for VEGF, wa...
متن کاملThe alternative complement pathway regulates pathological angiogenesis in the retina.
A defining feature in proliferative retinopathies is the formation of pathological neovessels. In these diseases, the balance between neovessel formation and regression determines blindness, making the modulation of neovessel growth highly desirable. The role of the immune system in these retinopathies is of increasing interest, but it is not completely understood. We investigated the role of t...
